\nLYNPARZA (olaparib) is a first-in-class PARP inhibitor and the first targeted treatment to block DNA damage response (DDR) in cells\/tumors harboring a deficiency in homologous recombination repair, such as mutations in BRCA1 and\/or BRCA2. Inhibition of PARP with LYNPARZA leads to the trapping of PARP bound to DNA single-strand breaks, stalling of replication forks, their collapse, and the generation of DNA double-strand breaks and cancer cell death. In the DDR pathways, LYNPARZA is being tested in a range of PARP-dependent tumor types with defects and dependencies.\n<\/p>\n

\nLYNPRAZA is indicated for the maintenance treatment of adult patients with deleterious or suspected deleterious gBRCAm metastatic pancreatic adenocarcinoma whose disease has not progressed on at least 16 weeks of a first-line platinum-based chemotherapy regimen.\n<\/p>\n

Olaparib has been shown to inhibit the growth of select tumor cell lines in vitro and decrease tumor growth in mouse xenograft models of human cancer, both as monotherapy or following platinum-based chemotherapy.\n<\/p>\n

\nIncreased cytotoxicity and antitumor activity following treatment with olaparib were noted in cell lines and mouse tumor models with deficiencies in BRCA1\/2, ATM, or other genes involved in the homologous recombination repair (HRR) of DNA damage and correlated with platinum response. In vitro studies have shown that olaparib-induced cytotoxicity may involve inhibition of PARP enzymatic activity and increased formation of PARP-DNA complexes, resulting in DNA damage and cancer cell death.\n<\/p>\n
